2.15.4 In-gel Fluorescence Visualization

The reactions were terminated by the addition of ice-cold acetone (4 volumes), placed at -20 °C for overnight and centrifuged at 6000×g for 5 min at 4 °C to precipitate proteins. The supernatant from the samples was discarded. The protein pellets were washed with ice-cold methanol once, air-dried for ~5 min, resuspended in 20 μL of loading buffer with 50 mM Dithiothreitol (DTT), heated at 85 °C for 8 min, and resolved by SDS-PAGE. The labeled proteins were visualized by scanning the gel on a Typhoon 9410 variable mode imager (excitation 532 nm, emission 580 nm).
